WARNING: Rogue Launched Trope. Edits must be edited to this draft, and wicks must be dewicked. Reverting to pre-launched state for scheduled unlaunching.
Supertrope page for tropes which link character traits to music, either diagetic or non-diagetic.
A great way to quickly establish a character's traits, or show a character's role, is to show them playing an instrument, as instruments and the musicians that play them are often shown to be similar.
For example, violins are often seen as sophisticated or melancholy. Trombones with their potential for physical comedy are seen as goofy. And anyone who plays the Accordion is either a dork or has a thing for Paris.

All Drummers Are Animals
Dumb and Drummer
Elegant Classical Musician
Harp of Femininity
Magical Flutist
Nobody Loves the Bassist
Nostalgic Musicbox
Ominous Pipe Organ
Sexophone
Compare with AM/FM Characterization, in which an individual song is used to establish a character.
The description for Five-Man Band associates an instrument with each member, as if it was a literal band. The Lancer plays the lead guitar to accompany The Leader's singing, The Smart Guy plays the keyboard, The Big Guy plays the drums, while The Chick plays the tambourine and provides vocals.

Sakamichi No Apollon is about a teenage boy in the 1960s who befriends another boy who gets him into jazz:
Kaoru had a Friendless Background and is a studious, intelligent person who plays classical piano.
Sentarou at first glance is a delinquent who gets into fights, but he is really a Gentle Giant. He plays the drums and likes jazz.
Junichi is a free-spirited Big Brother Mentor who also likes jazz and plays a trumpet.

In Back to the Future, Marty Mc Fly plays the electric guitar throughout the film. This helps establish his laid-back and semi-slacker personality.

One Piece: Brook, the musician of Straw Hat Pirates, initially plays the violin and sometimes piano to emphasize his relatively classy background and personality (read: being a musician in another pirate crew and, nowadays, is slightly insane). After the time skip, he switches to playing guitars as "Soul King" which reflects his more lively personality.
