A character is revealed to be... black. Or Asian. Or Jewish. Or any ethnicity that isn't the same as that of the majority of the target audience.
The other characters are surprised or shocked. More importantly, the audience is expected to be surprised as well. Ideally, the character will need to keep covered up before the reveal or have non-obvious features. Otherwise the trope provides comedy, possibly intentional.
This trope can be played in several ways. The fact that the character is not what the audience would assume can be used for a cheap shock effect. Sometimes, the fact that the other characters are surprised is parodied. Alternatively, the audience may be called out for assuming that all characters have a certain ethnicity unless otherwise specified.
Interestingly, if the character didn't know about their own ancestry beforehand, The Reveal often will trigger (at least in more fanciful works) a major personality overhaul. The character will now be fully conscious of their "blood" and begin identifying as an ethnic minority and seeing the world in a much different light. Or they might be unimpressed and attempt to play down their pedigree and become annoyed if anyone brings it up as something supposedly important.
If the story contains An Aesop, it's usually that all races are equal.
In these melting-pot days in western countries (particularly America), it is on the way to being a Discredited Trope. However, many non-western countries remain near-ethnostates and such a reveal may still be rare and shocking. Compare Samus Is a Girl and Gender-Concealing Writing. Contrast But Not Too Foreign where a seemingly foreign character is revealed to be at least partially a member of the target audience's nationality. Compare and contrast Half-Human Hybrids, where a "human" character is revealed to be partially an animal or a mythical creature.
Can overlap with Informed Judaism (especially in a Hanukkah Episode) and You Know I'm Black, Right?. May also overlap with Mistaken Ethnicity if people had assumed their heritage was something else.

The Simpsons
There are some ethnic reveals in the Simpson family history. Homer reveals to Lisa that they have a Native American ancestor (much to her chagrin, as she had made up being part Indian for a report and then confessed that she wasn't). Another reveal is Lisa finding out she's 1/64 black descended from a runaway slave named Virgil (which Grandpa knew all along but lied about because he's Suddenly Racist), though the credibility of this claim is debatable considering the fact that The Simpsons Uncensored Family Album shows the Simpsons' family tree, which makes no mention of Virgil, instead a character named Garwood Simpson taking his place (though the same tree does confirm that the Simpsons have Native American ancestry; specifically, Garwood's father Rupert married a woman named Winifred Running Goat).
Principal Skinner's Armenian ancestry came out in an episode in which it is revealed that he (partly) lied about his Vietnam War experiences - and that his real surname is "Tamzarian." Could be considered Fridge Brilliance, since Seymour Skinner had always been drawn with vaguely "foreign" features.
Moe Syzlak is apparently half Armenian and half monster. Although never mind that he seems to come from another European country each time it's brought up.
And of course, a 3rd season episode that revealed that Krusty is Jewish. His birth name is Herschel Krustofsky.

Who the Ice Gods that claimed to be her parents and brother in the "Ice goes home" Justice League International storyline were has yet to be explained. As seen in this fan cartoon, Ice's retcon has met with some fan bemusement. The retcon also comes dripping with Unfortunate Implications, as it's shown that her entire clan (save her parents) were con men and thieves, and the head of the clan, her grandfather, wanted to use her to further his schemes.

Arthur's Perfect Christmas: It's suddenly revealed that several of the characters (who are all presented as animals, so none of them really have a visible ethnicity) are actually from different ethnic backgrounds (The Brain is African-American, Francine and her family are Jewish, etc.). Francine seems to get it the hardest, as during the episode Muffy invites her to a Christmas party and gets really mad when she turns her down, thus giving the episode an excuse to talk about Hanukkah. The episode "Is That Kosher?" has her trying to get through the Yom Kippur fast.

Batman: Endgame, a comic that — like so many other stories featuring the Joker — is wildly speculative, floats the possibility that the Joker, whose early life and background are almost completely unknown, could, under his chemical-bleached skin (which is even suggested to be makeup at one point in the narrative), be fully or part American Indian, or at least an outsider reared in that culture from birth (perhaps a descendant of one of the English/Native "lost colonies" from elsewhere in America). Nothing explicit is ever spoken, but Dr. Paul Dekker, AKA Crazy Quilt, tells Batman that the Joker revealed to him that he is actually a 400-year-old being, rendered semi-immortal by a cell-reconstructing chemical called dionesium that he found in a cave beneath Gotham City centuries ago. Crazy Quilt even shows Batman one of his mad paintings, which depicts the Joker standing on a bluff in colonial dress, already waiting when the first European sailing ship is arriving in Gotham Harbor. Part of the story is true: dionesium is indeed real, and there is a cave beneath Gotham that contains a pool of it. But Batman doubts all the other details from the beginning - and, after questioning Uriah Boone, a resurrected assassin for the Court of Owls society and the only person confirmed to have been alive in Gotham in the 17th century, concludes from Boone's body language that his claim that the whole story is true is a lie. Indeed, the comic ultimately suggests that the tale is merely one more colorful fabrication on the Joker's part — although the last two pages of the comic subtly hint otherwise. However, the theory that the Joker has Native ancestry is pretty plausible when you consider that the only other possible interpretation of Dekker's account is that he's not human.

In Homestuck, once Jane enters "Trickster Mode", her skin goes from just being generic white (like all the other characters) to very obviously Caucasian. This was played for laughs by Jane declaring that she felt so… "CAUCASIAN!", but it was retconned to "PEACHY!" shortly afterwards because a number of dickwads decided to use it as an excuse to be dicks to POC fans.

Eventually, it turns out that he's ethnically black, and can appear so through use of make-up. At one point, Manhunter (Kate Spencer) tries to guilt-trip him into getting involved in a border dispute by claiming he has "forgotten what his skin color is". Green Lantern: Judd Winick, the writer who established Ice's Romani heritage, previously declared that Kyle Rayner's father was a Mexican-American named Gabriel Vasquez rather than an Irish-American named Aaron Rayner, with Aaron retconned into being an alias he used while undercover. This was further solidified after the New 52 reboot, which established that Kyle was aware of his Latino ancestry from the very beginning, with Omega Men even depicting him as a fluent Spanish speaker. This is a far cry from Kyle's early appearances, which instead placed a great deal of emphasis on his Irish heritage, with him and his mother Maura sometimes speaking in Gaelic to each other.

During the period where James Rhodes filled in for Tony Stark as Iron Man, he gets captured by the Mandarin. Dick Grayson, the first Robin and later Nightwing, was revealed to be of Romani descent decades after his initial introduction. Interestingly, in one interview about Batman Forever (1995), Chris O'Donnell referred to Grayson as "a gypsy lad"; many readers probably thought he was just being metaphorical.
Batman: Endgame, a comic that — like so many other stories featuring the Joker — is wildly speculative, floats the possibility that the Joker, whose early life and background are almost completely unknown, could, under his chemical-bleached skin (which is even suggested to be makeup at one point in the narrative), be fully or part American Indian, or at least an outsider reared in that culture from birth (perhaps a descendant of one of the English/Native "lost colonies" from elsewhere in America). Nothing explicit is ever spoken, but Dr. Paul Dekker, AKA Crazy Quilt, tells Batman that the Joker revealed to him that he is actually a 400-year-old being, rendered semi-immortal by a cell-reconstructing chemical called dionesium that he found in a cave beneath Gotham City centuries ago. Crazy Quilt even shows Batman one of his mad paintings, which depicts the Joker standing on a bluff in colonial dress, already waiting when the first European sailing ship is arriving in Gotham Harbor. Part of the story is true: dionesium is indeed real, and there is a cave beneath Gotham that contains a pool of it. But Batman doubts all the other details from the beginning - and, after questioning Uriah Boone, a resurrected assassin for the Court of Owls society and the only person confirmed to have been alive in Gotham in the 17th century, concludes from Boone's body language that his claim that the whole story is true is a lie. Indeed, the comic ultimately suggests that the tale is merely one more colorful fabrication on the Joker's part — although the last two pages of the comic subtly hint otherwise. However, the theory that the Joker has Native ancestry is pretty plausible when you consider that the only other possible interpretation of Dekker's account is that he's not human.

Adam: Legend of the Blue Marvel: This is the entire premise of the series. Adam Brashear, better known as Blue Marvel, was a Superman Substitute active as a hero in the 1960s, until one day his mask was accidentally ripped during a battle, and it was revealed to the world that he was Black. Because this occurred at the height of the Civil Rights Movement, it predictably exploded and enflamed racial tensions. White people started hating and fearing him for being "Colored" and having powers, even when he was saving them, while Black people began labeling him a "Race Traitor" for not using his powers for their cause, especially those with more extremist views. Eventually, in order to prevent things from becoming chaotic, Blue Marvel was forced to permanently retire from superheroing by the Executive Order of the US President. In She-Hulk (2004), Spider-Man himself, who has an ever-present full facemask and a costume that covers every inch of his skin, is once asked to reveal why he thinks J. Notably, he attempts to use his race to trick people into following him by claiming his actions against Aquaman were in an attempt to make Atlantis a paradise for minorities all along. This is, naturally, a total lie he used to personal gain, which amusingly still leads to the aesop that all races are indeed equal: in that all of them are capable of producing jerks (further reinforced by the revelation that he suffers from a form of autism, which is played as a Freudian Excuse prompting Aquaman to later magically cure him...and the first thing Black Manta does afterwards is stab Aquaman and brag about having murdered his infant son).
